Yes, you can bus from Iguazu to Lima. It's going to be a long ride. If you don't have any visa issues with Paraguay (US citizens need a visa), you can go from Iguazu through Paraguay, and on to Salta. Or you can avoid Paraguay -- there are busses that goe from Iguazu to Salta bypassing Paraguay (going through Posadas/Corrientes, etc). Then you can catch a connection from Salta (or maybe Jujuy) on to Chile. An alternate route would be to go up through Bolivia. Either way, through Chile or Bolivia. You'll have to make a few connections, but can get to Lima easily. It is a long journey to do in one shot. Give extra time for any unexpected occurrences (I was delayed about 12 hours by an avalanche on the Pan American highway on my way to Lima).

You have two alternatives already, as Marisa outlined it. Either through Asuncion, Paraguay to Bolivia, or Argentina to Chile.
There is one more, if you, which might be easier, depending on your visa situation. From Iguazu to Pantanal, which is all in Brazil, and from there across Bolivia to Peru.

As you're crossing through the whole continent, you'll need enough time, to make your trip enjoyable. You don't want to spend all your time on busses, do you?

I agree. I'd put Iguazu Falls at or near the top of any "must see" list for South America, and would go out of my way to get to & from there to see it...BUT, Moniak's point is a great one: That part of South America is just too beautiful to be spent on a bus ride, and Marisa is right--it's one LONG bus ride.

I'd explore options for getting from Iguazu to elsewhere in Brasil or Argentina from which a more direct, less time-consuming means might be possible. I might even look into blowing some cash or freq. flier miles towards a cheap flight within the continent.

I have been looking at doing something similar, albeit stopping of for a day or 2 at each change over.
My plan would be (highly liable to change) this:
Bus from Foz-Corumba
over the border to Corumba and on the Death train thru Bolivia to Santa Cruz,
Bus from Santa Cruz - La Paz,
Bus La Paz to Cusco,
Bus Cusco - Lima.
